Effects of hydrostatic pressure on the osteogenic differentiation of hPDLSCs

Abstract:Abstract] AIM:To investigate the effect of hydrostatic pressure on the osteogenic differentiation of human periodontal membrane stem cells (hPDLSCs).METHODS:HPDLSCs were continuously treated with 20,100 and 200 kPa hydrostatic pressure for 6 h.Cells without hydrostatic pressure treatment served as the controls.The expression of receptor activator nuclear factor kB ligand (RANKL)and osteoprotegerin (OPG)was assessed by quantitative real-time PCR and Western blot.RESULTS:The 20 kPa pressure significantly increased OPG expression in HPDLSCs (P <0.05),it showed no significant effects on RANKL expression (P >0.05),thus decreased RANKL/OPG ratio (P <0.05).When the pressure increased to 100 kPa,RANKL expression was up-regulated (P <0.05),OPG ex-pression was slightly decreased (P >0.05),leading to increase of RANKL/OPG ratio (P <0.05).200 kPa pressure reduced both RANKL and OPG expression,RANKL showed more reduction (P <0.05),therefore RANKL/OPG ratio was decreased (P <0.05).CONCLUSION:Hydrostatic pressure affect the expression of both RANKL and OPG.
