Effect of hypoxia preconditioning on the proliferation and angiogenic factor expression of human dental pulp stem cells

Abstract:AIM:To investigate the effects of hypoxia preconditioning on the proliferation and VEGF expres-sion of human dental pulp stem cells (hDPSCs).METHODS:hDPSCs were cultured under normoxia (210 mL/L O2 )and hypoxia (30 mL/L O2 )respectively for 72 h.Cell surface molecules were examined by flow cytometry.MTT method was used to observe cell proliferation and Live /Dead assay was used to observe the cell viability,qRT-PCR and ELISA were performed to measure the expression of VEGF and SDF-1.RESULTS:Strong positive expression of CD29,CD44 and CD90 was found in hDPSCs.CD34,CD45 and CD31 were negative in the cells.Dead cell was not found in both groups.hDPSCs in hypoxia group showed higher proliferation ability than that in normoxia group (P <0.05).Hypoxia significantly increased VEGF mRNA and protein expression (P <0.05),but showed no signifi-cant effect on SDF-1 mRNA expression.CONCLUSION:Hypoxia preconditioning has no influence on hDPSCs via-bility,but can promote the proliferation and VEGF expression of hDPSCs.
