Effect of toll-like receptor 4 on the proliferationin of human periodontal ligament cells

Abstract：AIM:To examine the effects of toll like receptor 4 (TLR4)on the proliferation of human perio-dontal ligament cells (hPDLCs).METHODS:Primary hPDLCs were cultured by tissue block enzymolytic method. Cells of Passege 3 were transfected with NC-shRNA and TLR4-shRNA respectively,cells without treatment served as the controls.72 hours after treatment,TLR4 mRNA expression was examined by real-time quantitative polymerase chain reaction (RT-PCR).MTT assay was used to test the proliferation of hPDLCs.RESULTS:TLR4 mRNA expres-sion in the TLR4-shRNA group was significantly decreased than that in other groups (P<0.05).MTT assay showed that the OD values of TLR4-shRNA group at 48 and 72 h were significantly lower than those of the NC-shRNA group or blank control group(P<0.05).CONCLUSION:TLR4 interference suppresses cell proliferation of hPDLCs.
