Effects of nicotine on the proliferation and interleukin-8 expression of human gingival fibroblasts

Abstract:AIM:To investigate the effects of nicotine on the proliferation and IL-8 expression of human gingival fibroblasts(HGFs).METHODS:HGFs were primarily cultured and identified.MTT method was used to ex-amine the viability of the cells stimulated by nicotine at the concentrations of 10-2000 μg/mL respectively for 72 h. ELISA was used to detect IL-8 concentration in the culture supernatant after 72 h stimulation .RESULTS:Nicotine dose-dependently inhibited the proliferation of HGFs (P<0.05).10-500μg/mL nicotine stimulated IL-8 secretion in HGFs,100μg/mL nicotine showed the most significant stimulating effect (P<0.05 ).CONCLUSION:Nicotine can inhibit the proliferation of HGFs and increase IL-8 production by HGFs,indicating that nicotine may play a role in the progression of periodontitis in tobacco users.
