AKT-Foxo3 a signaling pathway in regulation of MMP-20 gene expression in mouse ameloblasts
Abstract:AIM:To investigate the role of AKT-Foxo3a signaling pathway in matrix metalloproteinase-20 (MMP-20)gene expression in mouse ameloblasts.METHODS:AKT-Foxo3a and MMP-20 expression in mouse ameloblasts were examined by immunohistochemistry.Effects of AKT-Foxo3a signaling pathway on MMP-20 promoter were investigated by real-time quantitative RT-PCR and dual luciferase reporter gene assay system.RESULTS:Im-munohistochemical staining results showed that AKT and phosphorylated Foxo3a (P-Foxo3a)were both strongly ex-pressed in the nuclei of secretory ameloblasts.By RT-PCR,it was found that AKT inhibitor reduced the Foxo3a in-duced MMP-20 gene expression (P<0.05).Dual luciferase assay showed that AKT inhibitor decreased MMP-20 pro-moter activity (P<0.05).CONCLUSION:Foxo3a regulates MMP-20 gene expression through AKT-Foxo3a signa-ling pathway in mouse ameloblasts.
