Preliminary study of the biology of 3D cultured periodontal ligament stem cells

Abstract:AIM:To study the biology of periodontal ligament stem cells (PDLSCs)in 3D hanging drop culture. METHODS:PDLSCs were cultured by adherent (2D)culture and 3D hanging drop method(3D culture)at the densi-ty of 2.5 ×104 ,5 ×104 and 1 ×105 respectively.The morphology,viability and surface antigens of the cells were ex-amined by optical microscope,Live /Dead assay and flow cytometriy respectively.The mRNA expression of ALP and OPN was detected by qRT-PCR.Mineralized nodule was observed by alizarin red staining.RESULTS:3D cultured PDLSCs can form dense 3D spheroid.No central necrosis was found in the 3D spheroid culture by the cell density of 2.5 ×104 ,while necrosis was found in that by 5 ×104 and 1 ×105 .Surface markers of stem cells were expressed on PDLSCs without significant difference between 2D and 3D cultures.More mineralized materials,higher expression of ALP and OPN mRNA were found in 3D spheroid cultured cells (P<0.05).CONCLUSION:3D cultured can keep the stem cell biological characteristics of PDLSCs.
