Expression of transforming growth factor β1 and basic fibroblast growth factor in phenytoin-treated macrophages of rats

Abstract:AIM:To observe the effects of phenytoin (PHT) on expression of transforming growth factor (TGF-β1) and basic fibroblast growth factor ( bFGF) in macrophages of SD rat . METHODS:Different concentrations of PHT solution (5μg/mL, 25μg/mL, 50μg/mL) were added in the separated cell culture media, in which the cultured cells were identified by immunocytochemistry (IHC) method. The expression of TGF-β1 and bFGF was detected in the macrophages after 48 hours of PHT treatment by IHC staining. RESULTS:The PHT treatment induced a dose-dependent increase in expression of TGF-β1 and bFGF in macrophages. Even the lowest dose of 5μg/mL could significantly increased TGF-β1 and bFGF expression (P<0.05). CONCLUSION:PHT can activate macrophages and up-regulate the expression of TGF-β1 and bFGF.
