Culture and identification of the bone marrow mesenchymal stem cells of green fluorescent protein-transgenic mice

Abstract:AIM:To evaluate the differentiating ability of the bone marrow mesenchymal stem cells (BMMSCs) of green fluorescent protein (gfp)-gene mouse in vitro. METHODS:Bone marrow cells from femur of 3-4-week-old gfp mice were splited with ACK. Then the cells were cultured in adherent way. The adhered cells were observed by fluorescence microscope. Stem cell surface markers were identified by flow cytometer. After identification, the cells were induced to differentiate into bone and fat cells in vitro. RESULTS:Primary gfp-BMMSCs were CD29 positive, CD90 positive and CD45 negative as determined by flow cytometry. They could be sub-cultured for many passages without any decay in fluorescence intensity. After3 weeks and 2 weeks induction, the gfp-BMMSCs could differentiate into osteoblasts and fat cells respectively, and maintained initial fluorescence intensity . CONCLUSION:Adherent gfp-BMMSCs not only showed steady fluorescence, but also can proliferate and differentiate into different lines of cells. Gfp-BMMSCs are suitable tracing梒ells for studying the stem cell niche.
