Induced differentiation of ectomesenchymal stem cells to muscle cells

Abstract:AIM:To induce the differentiation of ectomesenchymal stem cells (EMSC) to muscle cells.METHODS: The EMSC were cultured in DMEM containing 5 μL/mL Dimethyl sulphoxide (DMSO). Reverse transcription-polymerase chain reaction(RT-PCR) and immunohistochemistry were used to identify the characteristics of the cells. The rat fibroblast was used as control. RESULTS: RT-PCR showed that after 7d induction, EMSCs expressed Myod mRNA. After 14d induction positive desmin staining was detect in EMSCs by IHC. The controll rat fibroblast did not express Myod mRNA and desmin after induction. CONCLUSION: The results indicated that the EMSC can be differentiated into muscle cells by the action of DMSO.
