A study of intracellular calcium ion release and its channel stimulated by calcium hydroxide in human pulp cells in vitro
Abstract:AIM: To investigate intracellular calcium ion release by Ca(OH)2 stimulus in human pulp cells in vitro and to detect a system of calcium channel. METHODS: The sixth generation of human pulp cells was cultured in RPMI1640 medium with Ca(OH)2, Ca(OH)2 and heparin, Ca(OH)2 and procaine. With the Fluo- 3/AM Probe loaded, the Ca(OH)2 was added to the medium and then the intracellular calcium level was detected by laser confocal microscope. RESULTS: Intracellular calcium concentration increased in pulp cells treated with Ca(OH)2, Ca(OH)2 and procaine, while it didn' t change in with Ca(OH)2 heparin treatment. CONCLUSION: Ca(OH)2 can stimulate the intracellular calcium release channel of inositol triphosphate receptor open in pulp cells in vitro and increase the level of intracellular calcium concentration.
