The expression of UBR5 and ITLN1 in epithelial ovarian cancer tissues and their impact on clinical prognosis of EOC

Abstract:Objective To investigate the expression of ubiquitin protein ligase E3 component N-recognition protein 5(UBR5)and internal agglutinin 1(ITLN1)in epithelial ovarian cancer(EOC)tissue,and to analyze the clinical prognostic value of their expression.Method A retrospective selection was conducted on 98 patients with EOC treated in the Obstet-rics and Gynecology Department of Inner Mongolia Autonomous Region People's Hospital from March 2018 to March 2020,with 50 normal ovarian tissues as the control group.Immunohistochemistry was used to detect the expression of UBR5 and ITLN1 in tissues.Spearman rank correlation analysis was used to investigate the correlation between UBR5 and ITLN1.Fol-low up for 3 years,Kaplan-Meier curves were used to compare the prognostic differences between UBR5,ITLN1 positive and negative EOC patients.Cox regression analysis was used to analyze the prognostic factors in EOC patients.Results The positivity rate of UBR5 in EOC cancer tissue was higher than that in normal ovarian tissue,and the positivity rate of ITLN1 was lower than that in normal ovarian tissue,with statistical significance(x2/P=40.694/<0.001,40.597/<0.001).There was a significant negative correlation between UBR5 and ITLN1 expression in EOC(rs=-0.759,P<0.001).The positivity rate of UBR5 was higher in FIGO stage Ⅲ,with lymph node metastasis than in stage Ⅰ-Ⅱ,without lymph node metastasis;while the positivity rate of ITLN1 was lower in FIGO stage Ⅲ,with lymph node metastasis than in stage Ⅰ-Ⅱ,without lymph node metastasis(x2/P=17.606/<0.001,20.103/<0.001;10.303/0.006,13.131/<0.001);The 3-year cumulative survival rate of EOC patients in the UBR5 positive group was lower than that in the negative group,while the 3-year cumulative sur-vival rate of EOC patients in the ITLN1 negative group was lower than that in the positive group,and the differences were statistically significant(x2/P=6.803/0.009,11.090/0.001).FIGO stage Ⅲ,lymph node metastasis,and UBR5 positivity were risk factors affecting the prognosis of EOC,while ITLN1 positivity was a protective factor[HR(95%CI)=1.467(1.203-1.788),1.374(1.141-1.656),1.510(1.055-2.161),0.667(0.470-0.947)].Conclusion UBR5 expression is upregulated and ITLN1 expression is downregulated in EOC,both of which are related to patient FIGO staging and lymph node metastasis,and are involved in promoting malignant progression of EOC tumors.They are new tumor markers for prognostic evaluation.
