Effect of lentivirus transfected low HDAC1 expression on the dry proliferation, migration and invasion of gastric cancer stem cells and its mechanism

Abstract:Objective To investigate the effect of HDAC1 expression on stemness proliferation, migration and inva-sion of human gastric cancer and the expression of stem cell related genes in human gastric cancer were observed by lentivirus transfected to low gastric cancer stem cells, and the effect and mechanism of HDAC1 low expression on the proliferation and differentiation of human gastric cancer stem cells were further explored. Methods From July 2017 to October 2017, at the cell laboratory of the General Hospital of the North China Petroleum Administration, the GCSCs were selected by flow cytome-try with Nanog as marker. The expression of HDAC1 in GCSCs was down-regulated by lentiviral vector and detected by real-time quantitative reverse transcription PCR (qRT PCR) and Western Blot. The difference of Proliferation and immigration be-tween two groups were evaluated by MTT assays and scratch test respectively; The difference of invasion between two groups were evaluated by transwell assays; The expression of stemness related genes was detected by qRT PCR and Western Blot. Re-sults The proliferation(t=7. 251, P=0. 001) and immigration (t=6. 813, P=0. 001)in the experimental group were ob-viously lower than control group. The expression of stemness related genes in the experimental group were obviously lower than control group(t=28. 306, P=0. 000; t=7. 288, P=0. 001; t=12. 841, P=0. 000). Conclusion The low expression of HDAC1 can decrease proliferation, differentiation and invasion in gastric cancer stem cells. The low expression of HDAC1 down-regulate the expression of BMI1, which was thought to be one of the mechanism that HDAC1 affect proliferation and differentiation in human gastric cancer stem cells.
