Effect of Danhong injection on cerebral edema and plasma levels of MMP-9, TIMP-1 and NF-κB in rats with experimental intracerebral hemorrhage

Abstract:Objective To observe the effect of Danhong Injection on cerebral edema and the levels of plasma MMP-9,TIMP-1 and NF-κB in rats with experimental intracerebral hemorrhage(ICH),and to explore the possible mechanisms to protect nerve function and reduce brain edema.Methods The experiment was conducted in Guangdong Academy of Traditional Chinese Medicine between March 2016 to December 2016,60 Male Wistar rats were divided into 3 groups randomly: sham operation control group,model group and danhong group.Established the intracerebral hemorrhage model with intracerebral injection of autologous arterial blood.The rats of the danhong group were given Danhong Injection by intraperitoneal injection(2 ml·kg-1·d-1),and those of the sham operation control group and model group were given physiological saline(2 ml·kg-1·d-1).The content of water of brain tissue and plasma MMP-9,TIMP-1 and NF-κB at 1d,3d,5d,7d were tested.Results Compared with the sham operation control group,the brain water content,plasma MMP-9,plasma NF-κB of the rats in model group were significantly higher,but plasma TIMP-1 lower;Compared with the model group,the brain water content,plasma MMP-9,plasma NF-κB of the rats in danhong group were lower significantly,but plasma TIMP-1 higher.Conclusion Danhong injection can protect the brain function and reduce brain edema in rats with intracerebral hemorrhage,and may be related to the regulation of MMP-9,TIMP-1 and NF-κB expression.
