Effects of liraglutide on contraction and relaxation of mesenteric artery in diabetic rats

Abstract:Objective To observe the effects of liraglutide on mesenteric vascular systolic and diastolic function of di -abetic rats.Methods Take male SD rats 20 of preparing diabetes animal model , take the mesenteric artery preparation of ar-terial rings, using DMT micro vascular tension determination is used to record the DM rats isolated mesenteric arterial tension change.To observe the effects of liraglutide on mesenteric arterial tension , detected in vascular tissues no and NOS content changes .Results Liraglutide can improve rat mesenteric arterial tension under basal conditions in vitro ;cumulative concen-trations of liraglutide (10 -8 -10 -4 mol /L) but also to pre constricted with phenylephrine ( PE) in diabetic rat vascular ring tension produced a concentration dependent vasodilation ;in addition, liraglutide for artery endothelium intact group diastolic rate was significantly higher than that of endothelium denuded group( t=10 .309, P =0.028);by ELISA, liraglutide groups of vascular tissue, the contents of NO and NOS were higher than K H solution control group ( t =16.802, P =0.017).Con-clusion Liraglutide of DM rat mesenteric artery with diastolic and systolic dual regulatory role , it pre contracted with PE DM rat mesenteric artery with concentration dependent relaxation effect , and is dependent on the presence of vascular endothelial and its mechanism may be by no and NOS of Shu Zhang resistance mesenteric artery .
