Expression of pancreatic tissue TLR4 and its inflammatory facot rsi n high fat diet induced insulin resistance in mice

Abstract:Objective To study the expression changes of Toll like receptor 4 ( TLR4 )and nuclear factor B kappa ( NF-κB) in pancreatic tissue, and to observe the level of inflammatory factors , to understand the relationship between insulin resistance and insulin resistance in mice induced by high fat diet, to provide basis for clinical treatment.Methods 160 male mice of C57BL/6 model were selected, the mice were divided into study group and control group according to the random num -ber table method,80 rats in each group, the control group was given normal diet, the study group was given high fat diet, each group of mice was divided into 8 subgroups according to the random number table method, 10 rats in each subgroup, the body weight, fasting blood glucose and insulin levels were measured in mice after 12 weeks of feeding, pancreatic tissue was taken from mice, hematoxylin eosin staining ( HE) , and then observe the morphology of islet cells, TLR4 and NF-κB protein expression in pancreatic tissue was detected by blot Western method, immunohistochemical method was used to detect the tumor necrosis factor a ( TNF-α) and interleukin 6 ( IL-6 ) expression level.Result Mouse model of insulin resistance was induced successfully, the body weight and fasting blood glucose of mice in the study group were increased with time, the differ-ence was statistically significant compared with the control group at 14 days and 21days( P <0.05),insulin levels increased&nbsp;from third days and reached a peak at seventh days , seventh days to forty second days and the control group had statistical sig-nificance compared with the control group ( P <0.05);High expression of NF-κB and TLR4 protein appeared in pancreatic tissue from seventh days, twenty first days to reach the peak, then decreased, the expression of TNF-αand IL-6 in pancreatic islet cells was also expressed at the same time.Conclusion High fat diet induces TLR4/NF-κB pathway activation in mice with insulin resistance, and the emergence of a large number of TNF-αand IL-6 expression.
