Expression of paxillin protein and mRNA in gastric carcinoma and analysis of bioinformatics

Abstract:Objective To investigate the expression and significance of paxillin protein and mRNA in normal gastric mucosa and gastric carcinoma, and to analyze the causes of the disorder.Methods Selected from January 2009 to December 2014, 37 cases of normal gastric mucosa and 199 cases of gastric carcinoma tissue in Wuhan University Renmin Hospital, using tissue microarray and immunohistochemistry to detect paxillin protein expression, and to analyze the relationship between the protein and clinical pathological parameters.The negative regulation of paxillin expression in miRNA protein was screened by bioinformatics methods.Results There was significant difference in paxillin protein in normal gastric mucosa tissue, the positive expression rate was 70.27%(26/37), which was significantly higher than that in gastric cancer tissue of 43.72%(87/199)(χ2 =8.814, P <0.05);along with decrease of gastric cancer cell's differentiation degree, paxillin protein expres-sion gradually decreased, the difference was statistically significant ( P <0.05).The positive expression in diffuse type gas-tric cancer rates was 31.25%(40/128),which was significantly lower than that in intestinal gastric type cancer 63.33%(38/60),and the difference was statistically significant (χ2 =17.322, P <0.05).In gastric cancer tissues of paxillin and ubiquit-in specific protease 10 (USP 10) expression was not found significantly correlation ( r =0.187, P >0.05).Paxillin mRNA expression spectrum data for normal mucosa:gastric cancer =1.04:1.Intestinal type:diffuse type gastric cancer =0.88:1, there were no obvious differences ( P >0.05), showed that paxillin expression down-regulation occurs at the post tran-scriptional level.miR-199a-5p and paxillin mRNA 3 'UTR terminal with a highly conserved binding site, and compared to nor-mal tissue showed high level expression in gastric cancer.Conclusion Paxillin protein expression and gastric carcinogenesis and tumor cell differentiation were closely related, the protein in gastric cancer down-regulation occurs mainly at the post tran-scriptional level, miR-199a-5p may is one of the factors in the negative regulation process.
