The therapeutic effect of memantine combined with donepezil on Alzheimer's disease

Abstract：Objective To observe the effect of donepezil hydrochloride combined with memantine on cognitive andlife ability of patients with Alzheimer's disease.Methods Forty-eight cases of Alzheimer's disease patients were randomly dividedinto 2 groups, 24 cases in each group.The control group was treated with donepezil hydrochloride , in observationgroup, patients were treated with donepezil hydrochloride combined with memantine treatment for 12 weeks, follow up werecarried out at every 4 weeks.Before and after treatment, Alzheimer disease assessment scale-cognitive component table(ADAS-cog), Neuropsychiatric Inventory (NPI), ability of daily life table (ADL), dementia behavior table (BEHAVEAD),Mini Mental Scale (MMSE) were used to assess these patients, and observed the occurrence of adverse reactions .Results There were no significant differences of these index score between the 2 groups before treatment ( P >0.05).After 12weeks of treatment, ADAS-cog score and NPI score and ADL score, BEHAVE-AD score were decreased in both of observationgroup and control group ( P <0.05), MMSE score were increased ( P <0.05), and the observation group’s each tablescores were improved more significantly than in the control group , the differences between the two groups were statistically sig -nificant ( P <0.05).There were 2 cases (8.33%) in the control group occurred with adverse reactions ; the observationgroup had no adverse reactions.Conclusion Donepezil hydrochloride combined with memantine has obviously improvementeffect in patients with Alzheimer's disease cognition and ability of daily living , which were better than single usage of donepezilhydrochloride.
