The study of the effect of ApoG2 combined with radiotherapy onautophagy and apoptosis of CNE 2 cells of nasopha-ryngeal carcinoma

Abstract:Objective To investigate the effect of ApoG2 combined with radiotherapy on CNE 2 cell autophagy and apoptosis in nasopharyngeal carcinoma ( NPC) .Methods Human NPC CNE were divided into blank control group ( only 2 cell lines with medium, without the addition of cells) and radiotherapy group, ApoG2 group, combination group (radiotherapy+ApoG2) , to determine the treatment sensitizing effects on cell growth of ApoG2+radiation by using the method of CCK was 8, Hoechst 33258 staining was used to observe the changes of cell apoptosis morphology of autophagy, observe the mor-phological changes of acridine orange staining;cell apoptosis rate changes were detected, Western bolt detection were used to detect the expression of Beclin1, Bcl 2 protein changes.Results There was significant difference of cell proliferation inhibi-tion rate between different radiation dose group ( P <0.01) , with the increase of radiation doses, nasopharyngeal carcinoma CNE 2 cell inhibition rate also increased;different ApoG2 drug concentration group among the cell proliferation inhibition rate also has significant difference ( P <0.01), with the increase of drug concentration, CNE inhibited 2 nasopharyngeal carcino-ma cell rate increased.4 groups’ CNE 2 cells apoptosis rate of revealed significant differences within groups ( F =140.2, P <0.01), apoptosis rate of combined group was significantly higher than the other 3 groups ( P <0.01).4 groups of Bcl 2 ( F =71.24, P <0.01), Beclin1 ( F =107.53, P <0.01) protein expression was statistically significant differences be-tween groups, the joint group’ s Bcl 2 protein expression was the lowest, the expression of Beclin1 protein was the highest. Conclusion ApoG2 combined with radiotherapy can through autophagy and apoptosis induction of nasopharyngeal carcinoma CNE 2 cell to inhibit the proliferation of CNE 2 cells.
