Influence of Seed Immersion and Sterilization on the Sterile System Construction and Germination of Allium Seeds

Abstract：In order to rapidly obtain sterile seedlings,four species of allium plants were taken as research objects,with their seeds serving as explants.The seeds were immersed in sterile water,then disinfected with sodium hypochlo-rite and alcohol,and then transferred to MS medium to obtain sterile seedlings.The results showed that the contami-nation rate of non-immersed seeds was higher.Immersion for 24h combined with cross use of multiple disinfectants was beneficial for the mass production of sterile seedlings.Immersion for 48h benefited the improvement of germina-tion rate,germination potential and germination index,as well as shortening of seed germination days.However,im-mersion for 48h combined with cross use of multiple disinfectants would reduce the seedling vigor rate and seed con-tamination rate.
