Biological Activities of Phenolic Compounds Extracted and Purified from Burdock Root

Abstract:To investigate the active ingredient contents,antioxidant,antibacterial,and hypolipidemic activities of phenolic compounds from burdock root,crude extract(CE)was obtained using 60% ethanol extraction.This was purified with AB-8 macroporous resin to yield F60,followed by further purification using D101 macroporous resin with 20%,40%,and 60%ethanol gradient elution to obtain F60-20,F60-40,and F60-60,respectively.Spectrophotometric methods determined the contents of total phenolics,total tannins,total flavonoids,and total phenolic acids.Antioxidant activity was evaluated through free radical scavenging(DPPH,ABTS+)and metal ion reduction(molybdenum,copper)assays.Antibacterial activity was assessed via microplate dilution method,while hypolipidemic activity was evaluated by lipase inhibition analysis.Experimental results indicated that F60-60 and F60-40 exhibited higher levels of total phenolics,tannins,flavonoids,and phenolic acids among all samples.These fractions demonstrated effective DPPH and ABTS+radical scavenging capacities,significant molybdenum/copper ion reduction capabilities,and notable lipase inhibition activity.CE showed inhibitory effects against Listeria monocytogenes;F60,F60-20,and F60-60 against Salmonella;and F60-40 against Bacillus cereus.
