Optimal Extraction of Ursolic Acid from Loquat Leaves by Box-Behnken Design and Response Surface Method

Abstract:Taking the ursolic acid yield of loquat leaves as an index and four factors including ethanol concentration ,liquid‐solid ratio ,microwave radiation time and ultrasonic time as the most influential param‐eters ,this paper established the multiple regression equation with predictive analysis to optimize the micro‐wave‐assisted ultrasonic extraction by Box‐Behnken design and response surface method .The result showed that the best yield of ursolic acid was up to 10 .443% with ethanol concentration at 78 .5% ,liquid‐solid ratio of 24 .4 mL/g ,ultrasonic time of 74 min and microwave radiation time of 204 s .Meanwhile ,the ursolic acid yield was significantly influenced by ethanol concentration ,microwave radiation time and ultra‐sonic time during the extraction process .The response surface was dramatically affected by ethanol concen‐tration ,liquid‐solid ratio and microwave radiation time .The interactions were prominent among ultrasonic time ,ethanol concentration and liquid‐solid ratio .
