In vitro culture,identification and biological characteristics of skeletal muscle satellite cells
Abstract:AIM: To establish a method for in vitro culture of skeletal muscle satellite cells(SMSCs) and to explore the biological characteristics of the cultured cells.METHODS: Rat skeletal muscle satellite cells were obtained with the two-step digestion using type I collagenase and trypsin and passaged following primary culture in vitro. Morphological observation of the cultured cells was conducted,with the growth curve drawn and the rate of myotube formation(RMF) calculated for assessing the proliferative and differentiation ability of the SM-SCs. Cell identification was performed with immunocytochemical staining.RESULTS: The cells obtained after the two-step digestion showed strong proliferative ability in the growth media and could form myotubes when cultured in differentiation media. The SMSCs were weakly positive while the myotubes strongly positive for immunocytochemical staining with α-sarcometric actin and myosin.SMSCs cultured in vitro have strong proliferative and dif-ferentiation ability and can be used for tissue engineering and gene therapy.
