Remote preconditioning induces neuroprotection against spinal cord ischemia-reperfusion injury in rabbits
Abstract:AIM:To investigate whether remote preconditioning with transient hind-limb ischemia could induce tolerance to transient spinal cord ischemia and reperfusion injury in rabbits.METHODS:Twenty male New Zealand white rabbits were randomly divided into two groups (n=10 each).Animals in the control group received sodium pentobarbital(SP)(30 mg/kg iv)1 hour before infra-renal abdominal aorta was occluded.Animals in remote preconditioning(RPC) group were anesthetized with SP(30 mg/kg iv) and received two cycles of 10 min hind-limb ischemia(inflating cuff to 26.6 kPa)and 10 min reperfusion at 30 min before spinal cord ischemia.Spinal cord ischemia was induced by occluding infra-renal abdominal aorta for 20 min.Hind-limb motor function was determined with the Tarlov criteria(in which 4 is normal and 0 is paraplegia) at 4 hours,8 hours,2 hours,24 hours and 48 hours after reperfusion.All animals were sacrificed at 48 h after reperfusion and the spinal cords (L5-7)were removed immediately for histopathologic study.RESULTS:All the animals survived the experiment.The neurological function in RPC group was better than that in control group at 48 hours after reperfusion(P=0.000).The number of normal neurons in anterior horn of spinal cord of RPC group was more than that of control group at 48 hours after reperfusion(P=0.000) and there was a strong correlation between the neurological function scores and the number of normal neurons in anterior horn of spinal cords(r=0.868,P< 0.01). CONCLUSION:Remote preconditioning with transient hind-limb ischemia induces neuroprotection against spinal cord ischemia-reperfusion injury in rabbits.
