Bioactivity of anti- infectious tissue- engineered bone
Abstract:Aim To explore new way of anti- infectious tissue- engineered bone during primary recovery in treating infectious fracture and bony defects. Methods Eighty- four male Kunming mice weighted from 24 to 27 g were prepared.The muscle bags inside right thigh of thirty mice were implanted rifampin tissue- engineered bone for observing characteristics of drug release in vivo.Another fifty- four mice were divided randomly into A,B and C group according to random number method(eighteen mice in each group).The mice in A group were implanted with rifampin tissue- engineered bone,those in B group were implanted with simple tissue- engineered bone and those in C group were implanted with bovine spongy bone.Then the osteoinductive ability of those were observed.Results ① The rifampin concentration(1.8 mg/L) in surrounding soft tissue of bone granulation was higher than minimal isorrheic concentration(MIC) of staphylococcus aureus(0.008 to 0.06 mg/L) within 21 days after using this material and the blood drug concentration of mice was lower.② There was no significant difference in the osteoproductive ability between this material and simple tissue- engineered bone(P >0.05).Which all had better effects than that of simple bovine spongy carrier(P< 0.001).Measurement of alkaline phosphatease(μ mol/s@ kg) showed that it was 43± 12 in A group, 48± 10 in B group and 22± 5 in C group(F=7.56,P< 0.01) on 7th day and 65.3± 3.2 in A group, 63.0± 2.8 in B group and 18.2± 2.7 in C group(F=34.86,P< 0.01) on 14th day,and 122± 20 in A group, 133± 12 in B group and 27± 7 in C group(F=53.86,P< 0.01) on 28th day.Conclusion Anti- infectious tissue- engineered bone with some dose has high osteoinductive ability and anti- infectious activity with little blood drug concentration.
