Relationship between the susceptibility to sporadic Alzheimer's disease and the gene frequency of leukocyte antigen
Abstract:Aim To investigate the relationship between the susceptibility to sporadic Alzheimer's disease(SAD) and the gene frequency of HLA- DR2,DRB1* 04 and DRB1* 09 and to reveal the pathogenesis of SAD from the point of view of molecular immunogenetics.Methods The gene frequency of HLA- DR2,DRB1* 04 and DRB1* 09 were detected separately by using sequence special primer- polymerase chain reaction(PCR- SSP) in 44 SAD patients and 45 healthy control subjects.Results The gene frequency of HLA- DR2 allele(28.4% ,OR=2.384,P< 0.05)was much higher in SAD patients than in normal subjects(17.8% ) while the gene frequency of HLA- DRB1* 04(5.7% vs 14.4% ,OR=0.313,P< 0.05)was much lower.There was no difference for the gene HLA- DRB1* 09 between the two groups.Conclusion HLA- DR2 allele may increase the risk of the onset of SAD while HLA- DRB1* 04 may play a protective role in the pathogenesis of SAD.There is no correlation between HLA- DRB1* 09 allele and the onset of SAD
