The effects of ischemia-reperfusion injury on life activities of skeletal muscle cells
Abstract:Aim To determine the effects of ischemia-reperfusion on activity of mitochondria by measurating the respiratory control ratio(RCR) of skeletal muscle on different conditions.Methods The respiratory control ratio of mitochondria isolated from ischemic and reperfused gastrocnemius with or without NSMA(3-nitro-N-methyl salicylamide) administered intraarterially were measured in ischemic and reperfused rat model produced by clamping right arterial iliaca communis.Results The respiratory control ratio of No.2,3,5,6 group decreased significantly than those of control group(P< 0.01),so did those of No.7 group(P< 0.05),while the respiratory control ratio of No.4 group was not apparently different from those of control group(P >0.05).In addition, the RCR of group with NSMA administered intraarterially increased significantly than those of corresponding group.Conclusion The mitochondria activity of skeletal muscle decreased markedly and the degree of oxidative phosphorylation coupling declined after being ischemic-reperfused.NSMA might affect the electron transference of respiratory chain and scavenge free radicals,which would improve the activity of mitochondria in skeletal muscle during the ischemic reperfusion.
