A Comparison of Two Approaches for Establishing a Unilateral Ureteral Obstruction Model in Mice

Abstract:Objective:To compare the effects of two approaches,transabdominal and retroperitoneal,for estab-lishing a unilateral ureteral obstruction(UUO)model in mice.Methods:Sixty-six mice were randomly assigned into three groups using a digital random number table:a normal control group(C group,6 mice),a transabdominal ap-proach group(P group,30 mice),and a retroperitoneal approach group(R group,30 mice).The C group under-went no surgical intervention,while the P group underwent left ureter ligation via a midline abdominal incision,and the R group underwent left ureter ligation through a retroperitoneal approach.The surgical time and incision length were compared between the P and R groups.Twenty-four hours post-operation,six mice from each of the C,P,and R groups were sacrificed.Blood levels of interleukin-6(IL-6)and tumor necrosis factor-alpha(TNF-α)were mea-sured and compared across the groups.Fourteen days later,the remaining mice were sacrificed,and differences in re-nal tissue periodic acid-Schiff(PAS)staining scores,overall survival rate,complication rate,modeling success rate,and learning curve were compared between the P and R groups.Results:The average incision length([1.55±0.28]cm)and average surgical time([5.13±0.78]min)in the R group were shorter than those in the P group([2.71±0.30]cm,[6.72±1.76]min),with statistically significant differences(P<0.001).The IL-6 concentration 24 hours post-surgery in the P group[(335.80±74.82)pg/ml]was higher than that in the R group[(513.4±119.7)pg/ml],with a statistically significant difference(P=0.02).Fourteen days post-operation,the survival rate and modeling suc-cess rate in the R group were both 100%,compared to 92%for both in the P group,with no significant differences(P=0.489 for both).The complication rate in the R group(4.17%)was lower than that in the P group(29.17%),with a statistically significant difference(P<0.05).The learning curve for the surgical modeling in the R group was shorter than that in the P group.Conclusion:Both approaches for establishing the unilateral ureteral obstruction mod-el in mice are safe and feasible.However,the retroperitoneal approach offers advantages such as smaller incisions,shorter surgical times,lower complication rates,and a shorter learning curve,making it a superior method for estab-lishing the UUO model in mice.
