Role of Nilestriol in Inhibiting Expression of Interleukin-6 Messenger Ribonucleic Acid (RNA) in Ovariectomized Rat Bone
Abstract:Objective: To investigate the effect of nilestriol on the expression of interleukin-6 mRNA in ovariectomized (OVX) rat bone. Methods:Thirty female Wistar rats with the age of 3 months were randomized into 3 groups with 10 rats each which included OVX, sham,and nilestriol groups. One week after ovariectomy, the nilestriol group was given nilestriol once a week for 13 weeks. All rats were killed 3 months postoperatively. Total RNA was isolated directly from bone,and the expression of IL-6 mRNA was detected using reverse transcription-polymerase chain reaction technique. Results: The level of IL-6 mR NA was lower in nilestriol group than that in OVX group (P < 0.01 ),and there was no significant difference between nilestriol and sham groups. Conclusion: IL-6 plays a role in the pathogenesis of posunenopsusal osteoporosis. Nilestriol can in hibit the expression of IL-6, reduce bone resorption, and maintain bone volume.
