Research on the Inhibition of Doxorubicin-resistant Cell Proliferation in Breast Cancer by Palbociclib Regulating CCNE1

Abstract:Objective To explore the mechanism by which palbociclib reverses doxorubicin-resistant human breast cancer(BC)cell line MCF7/ADR.Methods Different concentrations of palbociclib were added to the bc doxorubicin-resistant cell line MCF7/ADR cells for treatment to observe the effects of palbociclib on the proliferation and invasion of the bc doxorubicin-resistant cell line MCF7/ADR cells.The possible mechanism by which palbociclib inhibits the proliferation of doxorubicin-resistant cell lines MCF7/ADR cells was detected and verified by transcriptome sequencing(RNA-seq).Results The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay verified that the effective half-maximal inhibitory concentration(IC50)concentration of palbociclib for inhibiting the proliferation of the bc doxorubicin-resistant cell line MCF7/ADR was 3.8 μM.MTT,wound healing,Transwell,and 5-ethynyl-2'-deoxyuridine(EdU)assays collectively demonstrated that palbociclib can suppress its proliferation and invasion.Flow cytometry experiments confirmed that palbociclib could promote its apoptosis and inhibit its cell cycle at the G1 phase.Rna-seq assay revealed that palbociclib inhibits the proliferation of bc doxorubicin-resistant cell lines MCF7/ADR cells mainly by regulating the cell cycle and simultaneously inhibiting the cyclin E1(CCNE1).Overexpression of CCNE1 could partially reverse the proliferation and invasion ability of palbociclib against bc doxorubicin-resistant cell lines MCF7/ADR cells.Conclusion Palbociclib may affect the proliferation and invasion ability of BC doxorubicin-resistant cell line MCF7/ADR by regulating the expression of CCNE1.
