Mechanism of Curcumin Inhibits the Proliferation and Metastasis of MCF-7 Cells through the PI3K/AKT/mTOR Pathway

Abstract:Objective To observe the effects of curcumin on the proliferation,migration and metastasis of human breast cancer MCF-7 cells and explore the mechanism of the PI3K/AKT/mTOR signaling pathway.Methods The proliferation activity of human breast cancer MCF-7 cells was detected by cell counting kit-8(CCK-8)assay after intervention with different concentrations of curcumin.The invasion ability of MCF-7 cells was evaluated by Transwell assay.The phosphorylation status of p-AKT and p-mTOR in the signaling pathway of human breast cancer MCF-7 cells was analyzed by Western blotting.Results Compared with the blank control group,the proliferation and invasion inhibition ability of human breast cancer MCF-7 cells in different concentrations of curcumin groups were significantly reduced,and the differences were statistically significant(P<0.05).Compared with the blank control group,curcumin could up-regulate the expression of E-cadherin in human breast cancer MCF-7 cells,down-regulate the expression of N-cadherin and Vimentin and inhibit the phosphorylation levels of p-AKT and p-mTOR(P<0.05),and this effect showed a significant dose-time dependent relationship.Conclusion Curcumin can inhibit the growth of human breast cancer MCF-7 cells,weaken the invasion ability and increase the apoptosis rate of cells,and the mechanism may be related to the inhibition of the PI3K/AKT/mTOR signaling pathway by curcumin.
