Causal Relationship between Obesity and Heart Failure was Explored Based on A Two-Sample Mendelian Randomization Study

Abstract:Objective To explore the causal relationship between obesity and heart failure using the Mendelian randomization(MR)method.Methods A two-sample Mendelian randomization analysis method was adopted,with obesity-related genetic variations as instrumental variables and heart failure as the outcome indicator.Relevant datasets of genome-wide association studies were collected.Five methods,namely inverse variance weighting(IVW),MR-Egger,weighted median,weighted model,and simple model,were used to analyze the causal effects of obesity and heart failure.Results A total of 14 single nucleotide polymorphisms(SNPS)were screened out as instrumental variables.The results of the 5 MR Analysis methods were consistent,all indicating a causal relationship between obesity and heart failure.The IVW result:[OR=1.210,95%CI(1.140-1.284),P<0.01];Mr-egger result:[OR=1.267,95%CI(1.074-1.495),P<0.05];Simple mode result:[OR=1.185,95%CI(1.050-1.338),P<0.05];Weighted median result:[OR=1.193,95%CI(1.105-1.289),P<0.01];Weighted model result:[OR=1.097,95%CI(1.097-1.314),P<0.01].Conclusion Obesity may increase the risk of heart failure,which has certain guiding significance for the clinical prevention and treatment of heart failure.
