The Effects of Recombinant Human Endostatin Combined with TP and GP Regimens on Proliferation and Stemness Characteristics of Non-small Cell Lung Cancer Cells

Abstract：Objective To investigate the effects of recombinant human endostatin(YH-16)combined with TP(paclitaxel+cisplatin)and GP(gemcitabine+cisplatin)chemotherapy regimens on the proliferation and stemness characteristics of non-small cell lung cancer(NSCLC)cells.Methods Three NSCLC cell lines,namely A549,H1975 and H1299 were used.According to different drug uses,they were divided into the control group,the group using YH-16 alone(Group Y),the YH-16 combined with GP regimen group(Group YG),and the YH-16 combined with TP regimen group(Group YT).The cell survival rate of different drug combinations was evaluated by methods such as CCK-8 assay,flow cytometry detection,real-time polymerase chain reaction(PCR)and Western blotting.The influence of apoptosis rate and the expression of stem cell-related markers[rabbit anti-markers sex-determining region Y-box protein 2(Sox2),aldehyde dehydrogenase 1 family member A1(ALDH1A1)].Results Compared with the control group,the survival rates of NSCLC cell lines in group Y,group YT,and group YG were significantly lower,and the apoptosis rate of NSCLC cells was significantly higher.Meanwhile,compared with group Y,the survival rates of NSCLC cell lines in group YT and group YG were lower,the apoptosis rate of NSCLC cells was higher,and the survival rate of NSCLC cell lines in group YT was significantly lower than that in group YG.The apoptosis rate of NSCLC was significantly higher than that of the YG group,and the differences were statistically significant(P<0.05).The mRNA expression and protein levels of Sox2 and ALDH1A1 in the YT group,YG group and Y group were lower than those in the control group.Moreover,the YT group and YG group were lower than those in the Y group,and the YT group was lower than those in the YG group.The differences were all statistically significant(P<0.05).Conclusion Both the YH-16 combined with TP and GP regimens can effectively inhibit the proliferation of NSCLC cells and promote their apoptosis,while reducing their stemness characteristics.Among them,the YH-16 combined with TP regimen has a more significant effect.
