Human Blood Metabolome and Chronic Rhinosinusitis:Two Sample Mendelian Randomization Study

Abstract:Objective To evaluate the causal relationship between blood metabolomics and chronic rhinosinusitis(CRS).Methods Genetic associations with CRS were mainly derived from FinnGen(cases:8524;controls:167 849),while human blood metabolome were extracted from UK Biobank summary involving 115,078 participants.Heterogeneity testing,horizontal pleiotropy analysis,and sensitivity analysis were conducted using methods including inverse variance weighting(IVW),Mendelian randomization Egger regression(MR-Egger),weighted median,simple mode method,and Mendelian randomization of pleiotropy residuals and outlier(MR-PRESSO)to explore the causal relationship between blood metabolomics and CRS.Results The results of this study revealed a causal effect between 9 blood metabolomics and CRS.The Ratio of docosahexaenoic acid to total fatty acids showed an association with the risk of CRS[OR(95%CI):1.20(1.11,1.30),P=1.82×10-6].Additionally,the study revealed that eight blood metabolome contribute to lowering the incidence of CRS,particularly highlighting that omega-6 fatty acids notably diminish its frequency[OR(95%CI):0.79(0.72,0.88),P=5.92×10-6].There is no reverse causality effect of CRS on blood metabolome.Conclusion This study provides genetic evidence on the relationships of blood metabolome and CRS,offering valuable novel insights into the potential prevention and treatment of CRS.
