Preparation and Application of Broad-spectrum Antibody Against Human Papillomavirus L1 Capsid Protein

Abstract:Objective To establish a method for detecting human papillomavirus(HPV)L1 capsid protein in cervical cells.Methods A broad-spectrum monoclonal antibody that could react with multiple types of HPV L1 capsid proteins was obtained by immunizing mice and monoclonal antibody screening,a western blotting(WB)method to detect L1 capsid protein was established,the limit of its detection was verified,and the cervical exfoliated cells from patients with cervical lesions was tested.Results The antibody screened had the activity of reacting with at least 11 types of HPV L1 capsid protein,and could detect L1 capsid protein in cervical exfoliated cells,with detection limit of 0.4-3.7μg·mL-1.Conclusion The broad-spectrum antibody can be used for WB detection of HPV L1 capsid protein in exfoliated cells of cervical precancerous lesions.
