Experimental Studies on the Relationship between the Expression of Nuclear Factor-κB and Systemic Inlfammatory Response Syndrome and the Intervention Effect of Shenfu Injection

Abstract:ObjectiveTo investigate the relationship between nuclear factor _κB (NF _κB) and systemic inlfammatory response syndrome(SIRS), and the experimental intervention effects of Shenfu injection. MethodsThe experiment was designed into three groups: the normal control group, the lipopolysaccharide (LPS) injury group and the Shenfu injection (SF) treatment group. SIRS rat models were established by intravenous injection of lipopolysaccharide (LPS), and successfully screened by detecting toxins. After treated respevtively, the levels of blood mononuclear cell nuclear factor _κB(NF _κB), serum tumor necrosis factor_α (TNF _α), interleukin_ 6 (IL _ 6) at different time point were all detected by using euzymelinked immunosorbent assay(ELASA) technique. The pathological changes of lung and liver tissue were both observed.Results① The activity of NF _κB and TNF _α level in rat blood mononuclear cell were increased signiifcantly after injured by lipopolysaccharide(LPS) with the most obvious at 2 h (P< 0.05), then followed by a gradual decline.② The serum IL _ 6 concentration was continuely increased over time (P< 0.05).③ The results of pathology showed that lung tissue in LPS group appeared alveolar hemorrhage and edema, alveolar septum diffuse thickening , edema and inlfammatory cell inifltration. The liver tissue showed sinusoidal dilatation, congestion and liver cells focal necrosis. The above _ mentioned injuries of lung and liver tissues were all lessened notably in SF treatment group. <br> of NF _κB α and the levels of TNF _α and IL _ 6 of SF group were all decreased markedly (P< 0.05), the damages of lung and liver pathology were both alleviated. ConclusionNF _κB and other factors were involved in the occurrence and development of SIRS, and might lead to the damages of lung and liver. SF injection can reduce the activation of NF _κB, decrease the release of TNF _α and IL _ 6, alleviate the pathological damage of lung and liver, so it play a role of protection for patients with SIRS.
