Study on Warburg Effect in Bladder Tumor Cells and its Mechanism by Using Microlfuidic Chip Technology

Abstract:Objective To study Warburg effect in bladder tumor cells and its molecular mechanism.Methods T24 bladder tumor cells were conventionally cultured by using RPMI-1460 medium. Microlfuidic chip platform was used to mimic the bladder tumor cells’ microenvironment and assay the metabolites of aerobic glycolysis.The content of nicotinamide-adenine dinucleotide phosphate (NADPH) was detected by spectrophotometric method.ResultsThe microfluidic chip was designed and fabricated in order to assay the metabolites. The descending trend of NADPH content was observed within a certain time extent which suggested that NADPH was consumed in a reduced form, and the generating source by the pentose phosphate pathway was inhibited.ConclusionTumor cells mainly participate in the aerobic glycolysis pathway and partly the pentose phosphate pathway.
