A study on establishment of a stable animal model ofintrauterine adhesions

Abstract:Objective To establish a stable intrauterine adhesion animal model by using Phenol Mucilage. Methods 75 healthy,sexual mature S-D mice were chosen(15 male and 60 female).The female mice were ran-domly assigned togroups:30 in IUA group,15 in sham group and 15 in control group.A mouse model of intrauter-ine adhesion was established by usingPhenol mucilage. 10 days after the model establishment,5 mice in IUA group,7 in sham group and 7 in control group were killed.We observed the changes in the uterine cavity with na-ked eyes and classified them into different levels.Each uterine cavity was assessed by semi-quantitative fibrillation after HE staining and Masson staining. The changes of each group wasobserved and compared. The rest mice got natural pregnant. On the 10th day after pregnancy,we opened the uterus of the mice and counted the number of embryos as the fertility evaluation index and unveil how the intrauterine adhesion affectedthe embryo implantation. Results The uterus cavity in IUA group was narrow or even closed. Semi-quantitative score of intrauterine adhe-sion fibrosis was(6.6 ± 0. 98 points)for IUA group,0 for control group,and(-0.5 ± 1.91 points)for sham group,withsignificant statistical differences(P < 0.001). The difference between sham group and control group had no statistical difference(P > 0.05). The number of embryos was(0.2 ± 0.4)for IUA group,(6.3 ± 2.9)for sham group and(6.3 ± 1.8)for control group.The data difference was statistically countable(P<0.001),but the difference between sham group and control group had no statistical difference(P > 0.05). Conclusions Fertility in miceis remarkably reduced due to intrauterine adhesion. Phenol Mucilage can help successfully establish anani-mal model of intrauterine adhesion.
