Sodium valproate inhibit the expression of SCN3A gene

Abstract:Objective To determine the role of valproate in the expression of SCN3A and provide new evidence for the clinical treatment of epilepsy. Methods We treated the N1E-115 cells with sodium valproate and examine the expression of SCN3A mRNA. We also constructed the expression vector of the SCN3A gene promoter then transfected the vector into N1E-115 cells ,respectively detected the level of SCN3A mRNA and the expression level of firefly luciferase of N1E-115 cells with valproate treated and untreated. Results Valproate inhibited the expression of SCN3A significantly including the level of SCN3A mRNA and luciferase protein. Conclusion Valproate inhibits the seizures by reducing the expression of the SCN3A, which provides a new understanding of the treatment of epilepsy.
