Mendelian randomization study of immune cells and rosacea

Abstract:Objective To investigate the immune cells phenotypes associated with rosacea using Mendelian randomization(MR)analysis.Methods Based on the genome-wide association study database,a comprehensive exploration was carried out to identify the causal relationships between 731 immune cell phenotypes and the rosacea.Multiple regression models,including the Inverse Variance-Weighted(IVW)method,MR-Egger regression method,Simple Mode method,Weighted Median method,and Weighted Mode method,were employed for MR analysis.The robustness of the results was evaluated through sensitivity analysis,including heterogeneity test and horizontal pleiotropy test.Furthermore,multivariate Mendelian randomization analysis was conducted to correct the potential confounding effects of 6 positive immune cell phenotypes on the outcomes.Results Two-sample MR analysis showed that six immune cell phenotypes,namely Transitional%lymphocyte(OR=1.1742,95%CI 1.0173-1.3553),CD8dim NKT%T cell(OR=0.8670,95%CI:0.7726-0.9731),CD45 on CD14+monocyte(OR=0.8688,95%CI 0.7586-0.9952),CD62L-myeloid DC%DC(OR=0.9198,95%CI:0.8575-0.9866),Monocyte AC(OR=0.9428,95%CI:0.8911-0.9975),and CM DN(CD4-CD8-)AC(OR=0.8861,95%CI:0.7950-0.9877),had a causal relationship with the onset of rosacea.Reverse MR analysis did not find a reverse causal relationship between the six positive immune cell phenotypes and rosacea..Conclusion This study comprehensively evaluated the relationship between immune cell phenotypes and the risk of rosacea for the first time,providing an important theoretical basis for the immunotherapy strategies of rosacea.
