The role of miR-99a-3p in the effect of UVB on autophagy of B cells in systemic lupus erythematosus

Abstract:Objective TTo explore the potential mechanism of ultraviolet B(UVB)aggravating autophagy in systemic lupus erythematosus(SLE)patients and leading to disease progression,with the expectation of providing new ideas for the targeted therapy of SLE.Methods B lymphocytes(B cells)from normal human peripheral blood were isolated by immunomagnetic beads.The proportion of B cells was read by flow cytometry.Ball-1 and B cells were transfected with miR-99a-3p agomir,antagomir,and normal control(NC).After transfection,cells were irradiated with 50 mJ/cm2 of UVB light.The expression levels of miR-99a-3p,eukaryotic translation initiation factor 4E binding protein 1(EIF4EBP1),microtubule-associated protein 1 light chain 3 B(LC3B),and lysosome-associated membrane protein 2A(LAMP-2A)were detected using real-time quantitative PCR(RT-qPCR).Results Transfection of miR-99a-3p agomir reduced the expression of the target gene EIF4EBP1 and the autophagy-related genes LC3B and LAMP-2A.Transfection of antagomir increased the expression of EIF4EBP1,LC3B,and LAMP-2A.UVB radiation induced a decrease in the expression of miR-99a-3p in Ball-1 and B cells,while the expression of EIF4EBP1,LC3B,and LAMP-2A increased.Conclusion Strong ultraviolet radiation can reduce the expression of miR-99a-3p,aggravate autophagy in SLE patients,and lead to the progression of SLE.
