A research of the effect of fractionated microneedle radiofrequency on cockscombs—an animal model for port wine stains
Abstract:Objective To investigate the effect of fractionated microneedle radiofrequency(FMRF) on port wine stains(PWS) model cockscombs, and to explore the feasibility of treating PWS with FMRF. Methods One side of cockscombs were treated with FMRF, and the untreated side of cockscombs served as the self control. The radiofrequency was delivered to cockscombs with microneedle in the depth of 1.5 mm, with the radiofrequency pulse width of 1 000, 1 500, and 2 000 ms and the energy levels of 0, 10, 16, and 20 W, respectively. The cockscombs samples of each group were obtained on the 14th day and 28th day after being treated with FMRF. The reduced rate of cockscombs capillary and the ratio of collagen typeⅠ/Ⅲ were observed and calculated through HE staining and immunohistochemical staining. Results On the 14th day and 28th day after FMRF intervention, the reduced rate of the capillary number in the treatment groups were significant higher than the groups without RF treatment(all P<0.001). On the 28th day, to be compared with the other groups, the area percentage of collagen typeⅠ/Ⅲincerased significantly in the groups with the treatment parameters of 16 W/2 000 ms/1.5 mm, 20 W/1 000 ms/1.5 mm, 20 W/1500 ms/1.5 mm and 20 W/2 000ms/1.5 mm(P<0.001). Conclusion FMRF might effectively reduce the capillary number of the cockscombs through damaging the capillary network. With suitable radiofrequency energy levels and pulse width, the ratio of collagen typeⅠ/Ⅲ would remain stable in the recovered cockscombs. With excessive energy levels, the risk of cockscombs fibrosis would increase.
