Progress of pretreatment to enhance protoporphyrin Ⅸ accumulation and efficacy of topical photodynamic therapy in dermatology

Abstract:Topical photodynamic therapy (PDT) has been widely applied in dermatology.For neoplastic and non-neoplastic proliferative skin disease,PDT has many advantages such as prominent effect,high security and good cosmetic outcome.For the barrier function of the stratum corneum and the size,volume and infiltration depth of the lesion will affect ALA penetration,PDT is not ideal for thicker lesions.In order to optimize PDT effect,pretreatment of the skin prior to PDT which contains enhancement of photosensitizer permeation and improvement of enzyme activity of PpⅨ metabolism gradually get attention and development.The application of physical removal of skin barrier,chemical penetrant,iron chelators,Vitamin D3 analogs,etc in topical PDT will be briefly discussed in this review.
