NB-UVB regulation of HaCaT cells proliferation and apoptosis through Wnt/β-catenin signaling pathway: an experimental study
Abstract:Objective To investigate the effects of narrow band ultraviolet B(NB-UVB) regulation of HaCaT cells proliferation and apoptosis through Wnt/β-catenin signaling pathway.Methods Human HaCaT cell lines was adopted as the experimental subject and divided into four groups(blank control group,NB-UVB group,NB-UVB+BIO group,BIO group).Four methyl thiazolyl tetrazolium(MTT) assay was used to observe the cell proliferative capacity,flow cytometry(FCM) test was used to detect cell apoptosis,real-time quantitative PCR and Western-blotting were used to detect the expression levels of intracellular β-catenin,antiapoptotic gene survivin and proapoptotic gene caspease-3.Results Compared with the control group,the proliferation and viability of HaCaT cells were inhibited and cell apoptosis was induced in NB-UVB group,meanwhile decreased survivin expression and increased caspase-3 expression of HaCaT cells were also observed in NB-UVB group.Furthermore,these effects were found to be depend on the suppression of Wnt/β-catenin signaling pathway as pretreatment with the Wnt activator BIO markedly dampened the DHA-induced effects.Conclusion NB-UVB may inhibit skin HaCaT cells by suppressing Wnt/β-catenin signaling pathway.
