Study on the relationship between Mycoplasma genitalium infection and female mucopurulent cervicitis

Abstract:Objective To establish Taqman MGB real time fluorescence-based PCR assay for the detection of Mycoplasma genitalium(M.genitalium), and apply the method to evaluate the infection rate of M.genitalium in STD clinic patients who were suffering from cervicitis in our hospital, so as to find the correlation between M.genitalium infection and mucopurulent cervicitis(MPC). Methods One kind of Taqman MGB real time PCR assay was established for the detection of M.genitalium referring to the studies. A total of 74 patients with non-gonococcal, non-chlamydial MPC and 63 healthy female volunteers were recruited in our study. Cervical samples were collected and M.genitalium was detected by Taqman MGB real time PCR assay. Results The results indicated that the Taqman MGB real time PCR had good linear range, great repeatability and High sensitivity, the lowest detectable limit was 5 copies. The positive rates of M.genitalium in the MPC patients was 24.3% while none M.genitalium was found in the control group. Significant difference was found in the both two groups(P<0.001). In addition to infertility, several other factors including urethritis symptoms, sexual contact history, recurrent cervicitis and treatment history were associated with mycoplasma infection(P<0.001), and the positive rate of M.genitalium was 17.6%, 52.9%, 42.5% and 100% respectively. Conculsion The Taqman MGB real time fluorescence based PCR is a rapid and sensitive method for the quantitative and qualitative detection of M.genitalium. The prevalence of M.genitalium infection is higher in patients with non-gonococcal, non-chlamydial mucopurulent cervicitis attending STD clinic than that in normal population. Detection of M.genitalium has a great significance in guiding the treatment of recurrent cervicitis.
