Expressions of miR-320 and its downstream target gene survivin in psoriatic lesions
Abstract:Objective To measure the expressions of miR-320 and its downstream target gene survivin in psoriatic vulgaris lesions. Methods Tissue specimens were collected from lesions of 40 patients with psoriasis vulgaris and normal skin of 40 healthy human controls. Real-time PCR was performed to detect miR-320 and survivin mRNA expressions. Luciferase reporter assay was used to evaluate whether survivin is a direct target of miR-320. Statistical analysis was carried out by t-test for intergroup comparisons and by Pearson correlation analysis for the analysis of correlation between miR-320 and survivin expressions in psoriasis vulgaris lesions. Results Compared with the normal control skin, psoriasis vulgaris lesions showed significantly decreased mRNA expression level of miR-320 (0.561±0.1l, t=3.06, P<0.05), but increased mRNA expression level of survivin (2.034±0.26, t=3.35, P<0.05). The mRNA expression level of miR-320 showed a signiifcantly negative correlation with that of survivin (r2=0.634, P<0.05). Luciferase reporter assay showed that survivin was a direct target of miR-320. Conclusion MiR-320 might participate in the development of psoriatic lesions via targeting the regulation of its downstream target survivin.
