Effects of halometasone on expression of vitamin D receptor in human HaCaT keratinocytes
Abstract:ObjectiveTo investigate the effects of halometasone on the expression of vitamin D receptor(VDR) in human keratinocytes so as to evaluate the potential effects of halometasone on the therapeutic efifcacy of topical vitamin D3 analog in psoriasis.Methods HaCaT cells were treated by halometasone and calcipotriol with different concentrations for various durations. The cell proliferation was observed by CCK8 assay. Quantitative real-time PCR and Western blotting were used to detect the expression level of VDR mRNA and protein.ResultsHalometasone signiifcantly enhanced the inhibitory effects of calcipotriol on keratinocyte proliferation. Halometasone upregulated the expression of VDR at the level of mRNA and protein in a concentration-dependent manner.ConclusionThe upregulation of VDR expression by halometasone in keratinocytes may partly explain the phenomenon that halometasone could enhance the therapeutic efifcacy of topical vitamin D3 analog in the treatment of psoriasis.
