Effects of sirolimus on the autophagy in human skin ifbroblasts
Abstract:Objective To investigate the effects of sirolimus on the autophagy in human skin ifbroblasts(HSFs). Methods HSFs were isolated from the circumcised foreskin of a healthy male, and subjected to primary culture. After 4 to 10 passages, HSFs were incubated with sirolimus of 0, 0.1, 1.0, 10.0 mg/L for 24 h followed by the observation of autophagy. Acndine orange(AO) staining under lforescent microscope and transmission election microscope to detect autophagy, Western blot to detect the expressions of LC3-B and beclin1 in these skin fibroblasts. Results Intracellular acridine orange accumulation increased, and significant differences were observed between the 4 groups, beclin1 and LC3-B expression were up-regulated significantly compared to those untreated skin fibroblasts (all P<0.01). Conclusion The autophagy in HSFs can be upregulated by sirolimus.
