Causal association between breast augmentation and breast cancer risk:a bidirectional two-sample mendelian randomiza-tion study

Abstract:Objective To explore the impact of breast augmentation on the risk of breast cancer and its subtypes.Methods The exposure and outcome-related data in this study were derived from GWAS databases.The primary causal estimates were obtained using the Inverse Variance Weighting(IVW)method.Cochran's Q and Rucker's Q tests were employed to assess heterogeneity using both IVW and MR-Egger methods.Pleiotropy was examined with the Egger-intercept method,sensitivity analyses were conducted through leave-one-out tests,and F-values were calculated to evaluate the presence of weak instrument bias.Finally,MR PRESSO was used to validate the causal association between breast augmentation and the incidence of breast cancer and its subtypes.Results The results of the two-sample Mendelian Randomization study indicated that breast augmentation surgery did not increase the overall risk of breast cancer or the risk of estrogen receptor-negative breast cancer.However,breast augmentation surgery was associated with an increased risk of estrogen recep-tor-positive breast cancer.Conclusion This study elucidates the causal association between breast augmentation and the incidence of breast cancer and its subtypes.It holds clinical significance for detecting disease occurrence and understanding pathological changes following breast augmentation,providing theoretical support for further uncovering the molecular mechanisms of breast cancer and its subtypes.
