Long-chain non-coding LINC01535 inhibits the proliferation of pathological scar fibroblasts and induces apoptosis through the JAK/STAT3 pathway

Abstract:Objective To explore the effect of long-chain non-coding LINC01535 on pathological scar fibroblasts through the JAK/STAT3 pathway.Methods From February 2020 to May 2021,the human pathological scar fibroblasts was prepared in the Second Affiliated Hospital of Guangdong Medical University.The fibroblasts were divided into control group,empty group and LINC01535 group according to the difference of transfection of plasmid.CCK-8 method was used to detect survival rate of fibroblasts and flow cytometry was used to detect rate of fibroblast apoptosis.Hoechst 33258 staining was used to observe fibroblast morphology and RT-qPCR was used to detect LINC01535,JAK and STAT3 mRNA of fibroblasts.Western blot was used to detect JAK and STAT3 protein expression.Results The results of Hoechst 33258 staining showed that the nuclei of the control group and the empty group were normal and the fibroblast structure was intact.The nuclei of the human pathological scar fibroblasts in the LINC01535 group were stained deep and the inner core chromatin was constricted.Compared with the control group and the empty group,the rate of apoptosis,LINC01535 mRNA expression,JAK and STAT3 mRNA and protein expression levels of human pathological scar fibroblasts in the LINC01535 group were significantly increased,and the survival rate of fibroblasts was significantly reduced(P<0.05).The were no significant differences in all indexes between control group and empty group(P>0.05).Conclusion Overexpression of LINC01535 can target the activation of JAK/STAT3 signaling pathway and increase the expression levels of JAK and STAT3,thereby causing the survival of human pathological scar fibroblasts and inducing cell apoptosis.
