The effect of hyaluronidase on the histology of rabbit auricular cartilage

Abstract:Objective To explore the effect of hyaluronidase (HAase) on the glycosaminoglycan (GAGs) content of rabbit auricular cartilage in the early stage.Methods Different concentrations (75,150,300 U/ml) of HAase were injected under the rabbit auricle,while the same volume of normal saline was injected as a control.The cartilage of the control and treatment groups were harvested for Alicianblue staining and GAGs content detection at 3 and 7 days after injection.The GAGs content (including HA and sGAG) was detected by double antibody sandwich ABC-ELISA Assay and Dimethylmethylene Blue Assay (DMMB),respectively.The GAGs content in each group was analyzed.Results At the third and seventh days after injection,the content of HA and sGAG showed a generally a decreasing trend from the low HAase concentration group to the high HAase concentration group.On the 3rd day after injection,differences were apparent in the HA content between the control group and the medium (P<0.05) and high concentration group (P<0.05).The sGAG content was statistically significant between the control group and the other groups (the corresponding P value of the low,medium,and high concentration group was 0.003,0.003,and 0.001,respectively).On the 7th day after injection,the HA content was statistically significant between the control group and the other groups (P<0.05).For sGAG content,the difference could be seen in the high concentration group when compared with the control group (P<0.05) and the low concentration group (P<0.05).At the same concentration,the HA and GAGs content on the 7th day was higher than that on the 3rd day.The difference was statistically significant (P<0.05).Conclusion Early HAase injection can significantly reduce the content of GAGs (including HA and sGAG),and cause a histological change through the degradation of glycosaminoglycans.
